Cassette mutagenesis is a technique that allows the elimination of a gene segment flanked on both ends by restriction sites and replaced by a new restriction fragment containing base substitutions or deletions at specific sites. The consensus sequence is the ideal sequence of nucleotides or amino acids in which each position represents the most frequent base when comparing several sequences from the same region. DNA ligase is an enzyme that restores the broken phosphodiester bond (nick) in a double-stranded DNA strand and sometimes in an RNA strand. DNA polymerase sliding clamp is a protein complex of eukaryotic organisms that increases the processivity of DNA polymerase by binding to the catalytic subunit of DNA polymerase to DNA as DNA replication occurs at high speed.
